Structural studies on -carrageenan derived oligosaccharides

چکیده
Oligosaccharides were prepared through mild hydrochloric acid hydrolysis of -carrageenan from Kappaphycus striatum carrageenan. Three oligosaccharides were purified by strong-anion exchange high-performance chromatography. Their structure was elucidated using mass spectral and NMR data. Negative-ion electrospray ionization (ESI) mass spectra at different fragmentor voltages provided the molecular weight of the compounds and unraveled the fragmentation pattern of the -carrageenan oligosaccharides. 2D NMR techniques, including H–H COSY, H–H TOCSY and C–H HMQC, were performed to determine the structure of a trisulfated pentasaccharide. 1D NMR and ESIMS were used to determine the structures of a -carrageenan-derived pentasaccharide, heptasaccharide, and an undecasaccharide. All the oligosaccharides characterized have a 4-O-sulfo-D-galactopyranose residue at both the reducing and nonreducing ends. © 2002 Elsevier Science Ltd. All rights

Applications of Mass Spectrometry to Structural Analysis of Marine Oligosaccharides
Marine oligosaccharides have attracted increasing attention recently in developing potential drugs and biomaterials for their particular physical and chemical properties. However, the composition and sequence analysis of marine oligosaccharides are very challenging for their structural complexity and heterogeneity. Mass spectrometry (MS) has become an important technique for carbohydrate analys...
متن کاملStructural Analysis of -Carrageenan Sulfated Oligosaccharides by Positive Mode Nano-ESI- FTICR-MS and MS/MS by SORI-CID
Structural analysis of sulfated oligosaccharides from -carrageenan of up to ten residues (MW 2 kDa) was successfully carried out by positive mode nano-ESI-FTICR-MS together with MS/MS using sustained off-resonance irradiation-collision induced dissociation (SORI-CID). Glycosidic bond cleavage reactions via the Band Y-types of fragmentation were observed and enabled complete sequencing of the ol...
متن کاملDegradation of Marine Algae-Derived Carbohydrates by Bacteroidetes Isolated from Human Gut Microbiota
Carrageenan, agarose, and alginate are algae-derived undigested polysaccharides that have been used as food additives for hundreds of years. Fermentation of dietary carbohydrates of our food in the lower gut of humans is a critical process for the function and integrity of both the bacterial community and host cells. However, little is known about the fermentation of these three kinds of seawee...
متن کاملBinding affinities of vascular endothelial growth factor (VEGF) for heparin-derived oligosaccharides.
Heparin and HS (heparan sulfate) exert their wide range of biological activities by interacting with extracellular protein ligands. Among these important protein ligands are various angiogenic growth factors and cytokines. HS binding to VEGF (vascular endothelial growth factor) regulates multiple aspects of vascular development and function through its specific interaction with HS. Many studies...
متن کاملSulfated Oligosaccharides Mediate the Interaction between a Marine Red Alga and Its Green Algal Pathogenic Endophyte.
The endophytic green alga Acrochaete operculata completely colonizes the sporophytes of the red alga Chondrus crispus; however, it does not penetrate beyond the outer cell layers of the gametophytes. Given that the life cycle phases of C. crispus differ in the sulfation pattern of their extracellular matrix carrageenans, we investigated whether carra-geenan fragments could modulate parasite vir...
